The Dissolution enhancement of Meloxicam tablets from the synergistic effect of hydrotropic and sublimating agent

Abstract

Background and Objective: Drugs which fall into Class II of Biopharmaceutical Classification System suffer from poor aqueous solubility that limits the dissolution and oral bioavailability. The improvement of the dissolution rate of this class of drug is an important issue for enhancing their bioavailability and therapeutic efficacy. The objective of this study is to investigate the magnitude of enhancement of the dissolution rate of Class II drug “Meloxicam (MLX)” by using more than one solubility enhancement technique.

Methods: MLX-sodium benzoate solid dispersion at different ratios were prepared by kneading method to enhance the solubility, then MLX tablets were prepared by from solid dispersion mass and camphor as a sublimating agent in different concentrations. Compatibility of drug-excipients was studied using Fourier transforms infrared technique and the prepared tablets were evaluated for their physical properties and in vitro drug release behavior.

Results: The solubility of MLX was enhanced by 17-fold when it dispersed at 1:2 ratio with sodium benzoate. After 45 min a significant enhancement (P < 0.05) of the dissolution profile of F2, F3, and F4 tables was observed when compared with F1 tablets, that indicate the efficiency of solid dispersion in enhancing the dissolution of poor water soluble drugs, Moreover, after 5 min a significant change (P < 0.05) of the percentage drug release of F3 and F4 observed when they compared with F2 which result from the synergistic effect of the hydrotropic and sublimating agent to produce tablets with rapid disintegration and drug release. Conclusion: The dissolution studies indicate that sodium benzoate is an effective agent to enhance the percentage of drug release and the camphor is a valuable sublimating agent to achieve a rapid drug release and pharmacological action.
